Ligandrol (LGD-4033) is a non-steroidal oral SARM which binds AR with high affinity (Ki of ~1 nM) and selectivity. It elicits therapeutic effect of testosterone with an improved safety, tolerability and patient acceptance due to tissue-selective mechanism of action, and an oral route of administration.

In animal models this SARM demonstrates a robust selectivity for bone and muscle versus prostate and sebaceous glands, as well as anabolic and anti-resorptive activities in muscles and bones. Phase I clinical trial study showed Ligandrol to be safe, with a favourable pharacokinetic profile: the safety and tolerability have been established up to doses of 22mg; levels of haemoglobin, alanine aminotransferase, prostate-specific antigen, aspartate aminotransferase, and QT intervals are not altered significantly at any dose; the lean body mass is increased even during a short period without changes in prostate-specific antigen; it has a long elimination half-life as well as dose-proportional accumulation upon multiple dosing.

LGD-4033 administration can be associated with a dose-dependent suppression of total testosterone, SHBG (sex hormone-binding globulin), HDL (high density lipoprotein), cholesterol, and triglyceride levels.
